Enhancement of Production of Bioactive Compound and Antioxidant Activity in Inonotus obliquus by Bioconversion Process using Commercial Enzymes

Abstract
Background: Inonotus obliquus (Pers.:Fr.) Pilat is regarded as a functional food and medicinal resource due to the polyphenol content. However, the polyphenols in I. obliquus have a low absorption rate because they exist as a complex combined with proteins or carbohydrates. This study improves the production of free polyphenols through low molecularization in the bioconversion process using commercial enzymes including protease and carbohydrate-degrading enzymes.

Methods and Results: The total polyphenol content and radical scavenging activity of I. obliquus extract was significantly enhacned after bioconversion by commercial enzymes when compared to a hot-water extraction (p < 0.05). In addition, the I. obliquus extract was isolated from the bioconverted product using high performance liquid chromatography to investigate the main substances in the bioconverted products. The main substance was syringic acid at a concentration 73.6% (using Protamex) and 60.4% (using Flavourzyme) higher than the hot-water extract (p < 0.05).

Conclusions: The results confirm I. obliquus bioconverted using proteases contains a high antioxidant content when compared to the carbohydrate-degraded I. obliquus. Therefore, bioconverted I. obliquus using proteases should be developed as a high value-added raw material with excellent antioxidant potential for producing functional foods and medicines.
